when I connect to the VATSIM network using V-Pilot I can Hair everyone else but it does not go through to ATC

Like @Portalearth420 said, yes you do need to assign a PTT button.
I use Swift as my pilot client for Vatsim, I presume it would be the same for vPilot. For my joystick I assigned the trigger function as my PTT button. On my Honeycomb Alpha yoke I use a button on the left side for PTT.
