This can be a sign that your drive might be corrupted. You might want to run a System File Scan and check the health of the drive and the data on it.
Googled it and it appears to be caused by a corrupted file/drive…
I use these set of instructions built into Windows often to keep my Windows healthy. It will scan your operating system and compare it to the latest Windows Image (online) and restores the corrupted or altered files for you. Give it a try, it comes from Microsoft’s website.

My guess is,
Did you change the compatibility settings for the work around about the stutters, then you might have set it to read only in the checkbox.

Weekly I run the following fixes and repair options. You must have command prompt windows open at the administrator level. Doing it weekly keeps things in fine fettle. If you have large HDD the second one can take a while. For my 8TB drive it takes over a day to complete, as it’s USB.
SFC /scannow
once it finished I then run
chkdsk /f/r
Meridian, plug your USB externals to a USB3 or 3.1/3.2. I use an external box with 5 drives (2 are 8 TB in Raid 1) and I am getting the same performance as an internal SATA3.
Im not in the position to send a picture at this moment, but if you use the search bar on the taskbar from the desktop in Windows, you type in flightsimulator.exe. it should be in the top search, than select this with the right mouse click and select open location, than select with the right mouse click the file flightsimulator.exe and click properties, in this Boxtel did the workaround for the stuttering issue. (Well i dont know if you done this in the past because you didn’t mention it before, but that is why i guess).
In the box to the bottom i says, " read only" now make sure it is unchecked
If not than do that, so the update can fix by override.
If it is checked than it won’ update because permission will be denied by the system.

Hi all. I have exactly the same problem. This is a pain in the neck. Community folder is empty. Almost clean installation of the premium version. Last update of Benelux performed. The computer is with old items as from last October. MSFS is installed on C: Samsung SSD 970 EVO 1TB. I have done the full check of the drive with Samsung Magician. Done dism and sfc scannow i.a.w. with Microsoft support procedure. Done chkdsk c: /r. There are no other issues on the system. I used to run the sim several times every day before. The only thing that might have got in between the first failed run of the game is a windows update. I was surprised to find that this folder where is the *exe file is not accessible due to rights (I was not able to view it in Explorer). After giving rights I can access it, but still the error remains. I am not sure if I have done it correctly or maybe there are more folders with the same problem. There is no response on a ticket through Zendesk (third day). The only a little bit not common to find on my system is a virtual drive on A: from 64 GB RAM 32 is a virtual disk. Additional 8GB uses Asus software for cache speeding drives. From the 32 GB, 24 GB was set in MSFS to use as cache. I have updated Windows 10 and Nvidia and I have stopped NortonAV - no good. I am about to install MSFS once more. It is a pity they are not responding on the ticket. If anybody has got any other idea I can give it a try.
