I am assigning these buttons to my hotas and tried with a310, nothing happens

I just tried my A310 and the Heading & Altitude changes are being made from my Flight Stick.

Check your HOTAS buttons under Windows.

Correction #2
Correction

I had used my FSM to make the changes to heading & altitude.
It has encoders to spin.

I just tried to map one of my Flight Stick buttons to the “altitude increase”
on the A310 and it does work.

Mapped to “Increase Autopilot Reference Altitude”

Map to:
Turn Autopilot on/off = Toggle Autopilot Master
Increase Autopilot Reference Altitude
Decrease Autopilot Reference Altitude

Turn on Heading Mode on/off = Toggle Autopilot Heading Hold
Increase Heading Bug
Decrease Heading Bug

Can’t I change values without enabling hdg mode on?

No - if the aircraft is to respond to your HDG changes, it has to be in Autopilot ON and in HDG mode.

To answer his question:

He can change the heading bug without Heading mode on.
Also, the AP Reference Altitude.
