When reverting back to the non-beta version of MSFS 2024, I wonder if some of my setings will be lost, because I have to uninstall MSFS 2024 first.
Will I loose settings when doing so?
And if so, which files would I have to backup before uninstall - or would everything be stored in the cloud?
The most obvious one to backup is the usercfg.opt file. This stores your graphics settings and also where you have your community folder.
Everything else is stored online, as far as I know.
I have got into the habit of taking a copy of usercfg.opt every time I test some new settings out, in case i screw up badly.
It goes without saying that you need to keep the copy somewhere other than the default installation location or it will get wiped out.

All I can say is that every time I reinstalled MSFS24 it has created a vanilla usercfg.opt and I have to copy back my backup to replace it.
Better safe than sorry.
If you are using the Steam version: for that I can confirm that nothing gets lost. I went in and out of beta several times without issues
I would also make a backup of this folder:
%LOCALAPPDATA%\Packages\Microsoft.Limitless_8wekyb3d8bbwe\SystemAppData\wgs
The folder contains all your control bindings, and your logbook. Some users have reported in 2020 that reverting to non-beta has lead to either partial or total profile loss. That folder is your local copy of what is in the cloud, or at least a part of it.
If you are a Steam user I don’t know what the path would be.

i have different PCs with different graphic settings. So I guess these should be stored in this local file.
They are stored in the file mentioned.
They are also stored “in the cloud” and you should be prompted about using them during a reinstallation.

Nothing is lost.
But save your Community folder and any addons you have.
