If I override the autopilot in the V35 my moving the yoke, I hear the standard disconnect tone, but I cannot re-enable any autopilot functions.
Any suggestions appreciated.

I’m going to hate myself for saying this, but have you tried turning it off, and on again? The autopilot I mean.
LOL.. yeah. It seems manually overriding it turned it off permanently.
Not by my system to test, but are you retrimming the aircraft before you try to turn it back on? A lot of these don’t like to turn on when out of trim.
Thanks - I did, but into level flight configuration rather than climb, which I was in when I manually turned the aircraft. I’ll experiment a bit to see what happens.
Do you have a control binding set to disconnect autopilot (I can’t recall the exact naming, but it is not “Toggle Autopilot”)? I know with most of the aircraft if I press the button on my yoke to disconnect autopilot, I have to press it twice — once to disconnect and once again to put the AP into a state where it can be reenabled.
I wonder if the way Carenado programmed a yoke movement to cancel is using the same code as the disconnect button. If you set a control binding for the disconnect, try pressing that once after you’ve canceled the AP via yoke movement.
Thanks for the note - I have AP disconnect bound to a button on my joystick; I did try hitting it a few times trying to override whatever may have been going on, but nothing changed in terms of being able to re-activate the AP.
