Upon starting MSFS 2020, I go to the “World Map” page and plan the flight. But now the “Fly” button is no longer “highlighted” and I cannot start the flight! 
I am piloting the PMDG 737-700 and have been since its release.

No, I have not.
This is a guess. Something you have selected is either not available to or incompatible with MSFS. I would first try another airplane, then different airports. Like I said only a guess but could still help isolate the problem.

Have you tried with an empty Community folder?
This could be the result of an addon conflict.
Also try to delete your rolling cache as well!
